Current Scientific Research:
 
   Bioperine® used in this formulation is a patented form of piperine, an extract of black pepper. It has powerful thermogenic effects increasing the bio-availability of nutrients from the intestine by more then 60%. It stimulates the release of digestive enzymes, and increases the GI blood flow. It stimulates CNS activity in the brain stem, reverses respiratory depression, and has pain relieving action by reducing the level of Substance P, probably in the spinal cord. Piperine has a strong anti-inflammatory effect and increases the blood level of other anti-inflammatory agents (curcumin, vitamin C, etc.).
   Pepper has been known for thousands of years for its value as a spice which increases the taste and value of the food we eat. Today it accounts for about 35% of the world’s total spice trade. In traditional medicine it was valued for the treatment of intermittent fevers and stimulation of the gall bladder. It was also known as a nerve tonic. Together with ginger, it was known in Ayurvedic Medicine to “correct the three humours of the human organism.”  Pepper is known to taste “hot.” This is a manifestation of its physiological activity known to be antimicrobial, anti-parasitic, insecticidal, but also anesthetic. Piperine may deplete sensory nerves of the neurotransmitter called “Substance P”.
 
   Bioperine and respiratory illnesses: In a study involving 240 children, 78% of the participants reported significantly reduced frequency and severity of attacks. In another study of 20 pediatric patients, 100% showed significant clinical improvement.
   Bioperine as a bioavailability enhancer: The improved bioavailability of food nutrients and drugs is attributed to the active compound of pepper, piperine (Bioperine® is a 98% pure piperine extract). Piperine is thought to act in several ways:
  1. Binds to enzymes in the liver that bio-transform some drugs, thus increasing their bio-availability.
  2. Interacts with the process of oxidative phosphorilation, binds to enzymes causing glucuronidation, probably does not affect the elimination processes.
  3. In a study, the addition of 5mg of Bioperine to 15mg of beta-carotene, increased the level of carotene nearly twofold.
  4. In a separate study, serum levels of selenium in the experimental group were 30% higher than in the control group. The same bio-enhancing effect was seen in a study noting the 250% increased serum level of B6 when Bioperine was co-administered. A similar nearly twofold increase in serum levels was noted when the amounts of vitamin C were studied.
  5. In a study on Coenzyme-Q10, bioavailability was increased 30%. The expense of nutrients alone should make the increased bioavailability due to the presence of Bioperine worthwhile.
 
   Bioperine and its thermogenic effect: Piperine increases the blood circulation to the intestine. It is involved in the stimulation of secretion of catecholamine neuro-hormones leading to the secretion of epinephrine by the adrenal glands. Activation of beta-receptors by epinephrine increases levels of cyclic AMP, which in turn activates a host of enzymes resulting in biochemical reactions that produce heat energy.
   Piperine stimulates the thyroid gland and the action of the thyroid hormones with a stimulation of tissue oxygen uptake. Cellular studies showed an increase in the activity of mitochondrial enzymes, ATPases which release stored energy from ATP. Majeed makes the following statement: “Piperine has a thermogenic effect that can be characterized as “diet induced thermogenesis.” It represents an outcome of the digestive process and it is an indirect result of the absorption process and assimilation of nutrients. While Bioperine is thermogenic, it is also a thermonutrient – a substance that stimulates metabolism and thermogenesis in conjunction with increased nutrient absorption.”
 
   Bioperine and nerves: Bioperine is a stimulant and nervine. The stimulating activity has been postulated to occur in the brain stem controlling vital centers such as respiratory, circulation and the biological cycle of sleeping and waking. Piperine, for example, reverses the respiratory depression caused by morphine with the additional benefit of not reducing the pain relieving effect of the drug. Piperine increases pain relieving beta-endorphins in the brain. Endorphins dull the sense of pain and tide over the effects of trauma. They control, however, also the sense for hunger, thirst and our sex drive. Piperines belong to a class of substances that are anticonvulsant, antidepressant, and muscle relaxing. Mice treated with piperine showed an elevation of serotonin within an hour of substance administration. Piperine also reduces pain perception through reduction of nerve transmission of Substance P. 
 
   Bioperine and the gastro-intestinal tract:
  • Increases the amount of mucosal pancreatic secretions.
  • Reduces gastric acid secretions and ulcerations.
  • Increases glucose absorption. 
  • Interacts directly with the cells of the small intestine increasing their ability to absorb amino acids such as leucine, isoleucine and valine. The probable mechanism is stimulation of the gamma-glutamyl transpeptidase (GGT) enzyme. This cycle is important in the absorption of various other amino acids, other peptides and trace elements. The effect on GGT is extremely important in detoxification processes in the body. In related observations it was found that administration of piperine prevents depletion of the crucial glutathione probably in part by increasing the absorption of the sulfur containing amino acids methionine and cysteine. People with good health had reportedly higher concentrations of glutathione.
 
   Bioperine and anti-allergic and anti-inflammatory effects: Piperine has been shown to lower the level of liver lipid peroxides known to correlate positively with the degree of inflammation. Piperine lowers the level of mast cell degranulation.
 
   Ginger is a well known digestive, gall bladder, and liver stimulant. It is one of few herbal compounds that have a dual inhibitory action on the inflammatory arachidonic acid metabolism. It inhibits both the 5-lipoxygenase and the COX-2 pathways. In some studies gingerols were found to be more potent than indomethacin. Contains significant doses of melatonin. Other benefit categories are: antipyretic, cholesterol lowering, anti-ulcer activity, confirmed antiemetic and anti-nausea, antimicrobial and anti-parasitic activity.
 
   Enzyme Mix (DigeZyme, CereCallase): Enzymes have been added to the formulation to facilitate digestion of whole foods in the diet. Most people in the western world have some enzyme deficiency by the time they reach adulthood. DigeZyme - from a vegetarian source - facilitates the breakdown of fats, proteins and carbohydrates. It consists of a blend of amylases, cellulases, lactase, lipase, and neutral proteases. The inclusion of lactases facilitates milk digestion.
   CereCalase™ is an exclusive blend of hemicellulase, beta-glucanase, and phytase that is designed to synergistically disrupt the fibrous nature of plant cell walls and release trapped nutrients from plant materials. A large portion of the fibrous components of botanicals are composed of non-starch polysaccharides (NSPs). The primary NSPs found in the fibrous structure of plant cells include hemicelluloses and beta-glucans. Hemicellulose and beta-glucans can alter
the transit times of foods, bind digestive enzymes, and trap essential plant constituents. Enzymes capable of degrading these have been shown to improve the digestibility and nutrient profiles of plant foods. The enzyme mix allows optimal nutrition with a reduced amount of bloating and gas due to high fiber content.
 
   Trace Minerals have been included in this formulation because optimal absorption will not occur if mineral presence is low in the first place. Our soils are so depleted of trace minerals, and chemically based agriculture so further aggravates the problem, that a supplementation is required. The lack of trace nutrients is all the more vexing since practically no function in the body can progress properly without adequate amounts of trace minerals. Dosage and Administration:
   Take one capsule before a meal, twice daily. Enzymes Plus should be taken within 10-15 minutes before for best results. 
   To increase the bioavailability of a drug there does not need to be a direct time connection between administration of Enzymes Plus and the drug. There is, however, a time connection between food absorption and timing of Enzymes Plus.
Cautions:
   Piperine has been shown to enhance the bioavailability of several drugs used for the treatement of tuberculosis, cardiac diseases, infections, and asthma (phenitoin, antibiotics, rifampicin, theophyllin, propranolol, etc.). Enzymes Forte may affect drug blood levels up to three weeks after stopping the supplement. Consult your doctor if you are taking any medication requiring blood level needs to be monitored closely. Although the data is highly inconclusive, piperine may lower reproductive fertility. Stop if negative reactions occur after the first dose is taken. Do not take if allergies to any component are known.  Keep out of the reach of children. Do not take if pregnant or lactating.
